Lawyers for the 20-year-old Ukrainian ranked 29th made a last-gasp appeal to the Court for the Arbitration in Sport in Lausanne to allow her to play the Open.
The player had tested positive for a banned substance but said the incident must have been caused by cross contamination of some kind.
She was suspended by the International Tennis Federation but allowed to board one of the special charter flights for players heading to Melbourne and served out the 14-day quarantine.
Yastremska tested positive last November for the banned substance metabolite of mesterolone after testing negative a week earlier at a WTA tournament in Linz. .
The player will be unable to compete until a final decision/punishment in her case is made by the official anti-doping body.
